There is plate setting offset for the top corner (A1 well) which was incorrect in the template.

**Problem** Well location misaligned. Nunc 96 well plate 167008.

**Symptom**

- Field on view with the 20X objective falling outside the wells. Not a problem on the 10X objective.

- The plate model was checked. It matched the plate as option in the software dropdown, but not listed in the manual.

- The `Plate Alignment` steps were followed from page 56 of the manual
- Approximate centre of the TL well found by stepping in half way the top / bottom edge and the left / right. This was bit approx. Better to measure the FOV and step a known distance using the control in the `Fine tune` tab.

- This was repeated for TR and BL. There was a warning about a large correction being applied.
- Fields of view fall within the wells and assay ran.
